Abstract:
This study aimed to determine the changes of some kinematic parameters of ram spermatozoa in production of frozen sex-sorted semen from ram by BSA column. The ejaculetes were collected by artificial vagina method, then mixed, split of 6 equal parts and subjected to sexing by BSA column, cooling and freezing. Progressive motility (PM), non progressive motility (NPM) of spermatozoa, immotile sperms (IS) and values of speed including curve linear velocity (VCL), straight-line velocity (VSL) average path velocity (VAP), linearity index (LIN), straightness (STR) and oscillation index (WOB) after collection, sexing, cooling and thawing were measured by CASA. The results were statistically processed and compared. The ram semen sexing by BSA column affected insignificantly the kinematic parameters of the spermatozoa in upper layer (X fraction), but resulted in significant (P<0.05) change in PM, VCL and VAP of the spermatozoa in bottom layer (Y fraction). The cooling and the freezing decreased significantly (P<0.05) the sperm motility chracteristics in both fraction and resulted in clearly expressed drop in the average values of VCL and VAP for the Y fraction. The fraction with X spermatozoa was more resistant against negative effects of the freezing, compared to this with Y sepmatozoa. The obtained information can be used in improvement of the protocols for production of frozen sexed semen from rams.
